Features of the 6400 Visible and 6405 UV / Visible Spectrophotometers for a wide range of applications

Photometrics

Select this mode for the routine measurement of:

  • % Transmittance (0 – 199.9%)

  • Absorbance (-0.300 to 3.000A)

  • Concentration (range –300 to 9999)

  • Concentration by Factor (range 0.000 to 9999.99)

  • Unit selection (ppm, mg/L, g/L, %, M and blank)

 Spectrum

The 6400 & 6405 have full scanning capability with

  • 3 scan speeds (166, 400 and 1200 nm/minute)

  • 3 scan intervals (0.2, 1 and 5nm)

  • Baseline correction

  • Instant display of spectrum

  • Automatic axis scaling

  • Peak and valley data display

  • Hard copy of data/scan with printer accessory

  • 2 scans stored in memory

 Multi-wavelength analysis

For applications such as DNA/RNA composition @260/280nm, where difference and ratio analysis can be used quantitatively.

  • Simultaneous display of Absorbance or %T at two selected wavelengths.  Sequential calibration at each wavelength

  • Difference and ratio display (0 to 9999)

Kinetics

Software for Kinetics is included as standard. Temperature control is achieved with the Peltier cell accessory.

  • Lag time selection (0 – 9999 seconds)

  • Measurement time (10 – 9999 seconds)

  • Graphical display of data

  • Line of best fit between two user set points

  • Kinetics factor input and rate calculation

  • Statistical display gives initial absorbance, mean rate of absorbance change, kinetic activity value, and equation for line of best fit

  Quantitation

This mode is only available when the 8 position Multi-Cell Changer is fitted

  • Concentration calculation at a single wavelength from Absorbance or Transmittance readings

  • Concentration range, 0 – 9999, auto-ranging to a maximum of three decimal places

  • Automatic calibration sequence initiated via set-up menu

  • Up to 7 calibration points can be used

  • Choice of curve fitting methods:
            -Linear interpolation
            -Linear regression (line of best fit)
            -Line of best fit through zero

  • Print out of curve and numbered sample readings

  • 2 calibration curves stored in memory

GLP

Protocols are supported, by the following standard features:

  • User can set instrument number (up to 6 digits)

  • Time and date of last calibration

  • Print out states sample number, the reading and units of measure, wavelength of analysis, plus time and date

  • Stored scans along with concentration curves are stamped according to time and date.

Data Handling / Interfacing

Bi-directional RS232 Interface to allow remote operation, calibration and interrogation by an external PC.

  • Fixed baud rate at 1200 bits/second

  • Results can be downloaded by a single touch of the print key

  • Analog output (± 2000mV) is provided for use with a chart recorder

  • Output is proportional to the displayed reading (1mV per 0.1%T, 0.001A or 1 concentration unit)

Regulatory Compliance

  • The 6400 series are supported by traceable calibration standards, to provide the on-going accuracy required for Good Laboratory Practice (GLP) protocols.
